The X-LAB® System is designed to rapidly isolate mononuclear cells from various sources in a closed, sterile, semi-automated fashion without the need for density gradient media.

The X-LAB® System is a sedimentation-based process that quickly and reliably isolates mononuclear cells without the need for density gradient media or manual transfer steps. Traditional separation methods are open, manual processes that inherently introduce the risk of contamination as well as cell loss due to the multiple tedious processing steps. Contrary to these methods, the X-LAB System provides consistently high MNC isolation quickly without the dependence on highly skilled laboratory personnel and is therefore cost-effective and provides the standardization needed in a busy processing facility.

Working with the X-LAB® System

Cell Separation

The X-LAB® System is an automated blood separation and concentration system with a sterile fluid path. The system uses a combination of disposable and electromechanical components to deplete red blood cells from the initial blood product and provide an MNC-enriched output.

The X-LAB® Control Module employs highly accurate sensors to separate the blood product in the Disposable Cartridge’s three chambers (central processing chamber, red blood cell depletion chamber, and harvest chamber) for maximum separation of components without the use of density gradient media.

 

Process

Source material is transferred to the X-LAB® Disposable Cartridge via sterile welding or by a spike port. The 150-micron clot filter located on the input line allows for removal of clumps and promotes a single cell suspension for processing. After transfer, the input line is removed, then the Cartridge is attached to the X-LAB® Control Module, balanced with  balanced with another Disposable and Control Module set or an X-Counterweight and X-Balance Ring accessories, and the Control Module is activated prior to centrifugation.

The X-LAB® System is used in conjunction with a commercially available linkable centrifuge that allows for optimal component stratification and separation. During centrifugation, the blood product is stratified, then red blood cells are transferred into the Disposable Cartridge’s depletion chamber, and the MNC fraction is delivered to a harvest chamber.

The harvested cellular fraction can be removed from the Disposable Cartridge by sterile welding to a transfer bag or by syringe.
